Summary:Patchouli leaves contain essential oils that can be used as a component in making perfumes. Unfortunately, the cultivation of patchouli still faces issues related to the low yield of oil because patchouli is susceptible to viruses and bacteria. In vitro propagation using the TIS-Twin Flask bioreactor can increase metabolite production. This research aims to determine the influence of cultivating patchouli in the TIS-Twin Flask bioreactor and the effect of 10 µM methyl jasmonate elicitation on plant growth and metabolite accumulation, seedling survival rate, and oil yield. The results of this study show an increase in biomass amount by 122.907 ± 0.51%, a high growth rate of 0.502 ± 0.22 mm/day, and an RGR of 0.057 ± 0.007 ln(g)/day in cultures elicited with methyl jasmonate, while without elicitation, there was an increase in biomass amount by 75.037 ± 0.81%, a high growth rate of 0.167 ± 0.06 mm/day, and an RGR of 0.038 ± 0.015 ln(g)/day. Analysis conducted on the cultivation medium showed a decrease in sucrose levels, pH, and medium conductivity in both treatments compared to the initial conditions. The survivability rate from the cultivation of the TIS-Twin Flask reactor planted in soil was 100% with elicitation and 83.33% without methyl jasmonate elicitation. The patchouli oil produced through steam distillation from the cultivation of the TIS-Twin Flask reactor with and without elicitation was 9.09 ± 1.51% and 6.14 ± 1.39%, respectively. Cultivation of Pogostemon cablin in the TIS-Twin Flask bioreactor with 15-minute immersion intervals of 1 hour elicited with 10 µM methyl jasmonate can increase the growth and accumulation of patchouli oil in Pogostemon cablin leaves
