Summary:The patchouli plant (Pogostemon cablin Benth.) is a patchouli oil producing plant. Patchouli oil is composed of several secondary metabolites including terpenoids, flavonoids, alkaloids, steroids and phenylpropanoid glycosides. Thus, the oil has strong pharmacological activity and can be used as a raw material for aromatherapy, perfume, cosmetics, insecticides, and pharmaceuticals. The growing demand for patchouli oil in Indonesia has not been fully met because conventional cultivation is considered difficult to produce patchouli oil yields with good quality and high patchouli alcohol content. In vitro cultivation in a TIS-RITA® bioreactor can produce superior patchouli seedlings with similar characteristics, pathogen free and stress tolerant. In this study, in vitro cultivation of patchouli was performed in the TIS-RITA® bioreactor at a flow rate of 15.67 mL/min with the addition of 10 µM MeJA. MeJA elicitor acts as a self-defense response to induce genes involved in the patchouli alcohol biosynthesis pathway. The purpose of this study was to determine the effect of the addition of 10 µM MeJA elicitor on the growth and oil yield of patchouli when grown in vitro in the TIS-RITA® bioreactor. In the results of this study, there was an increase in growth with MeJA 10 µM treatment in the TIS RITA® bioreactor, which included the percentage of biomass increase of 42.265 ± 4.1%, the relative growth rate of 0.025 ± 0.004 g/day, the average growth rate of 0.031 ± 0.004 cm/day, and the percentage of shoot growth of 46.905 ± 4.2%. Meanwhile, the growth of the control was lower. The survival rate of patchouli in both control and treatment was 100%. Patchouli oil yield obtained by steam distillation in MeJA 10 µM treatment was higher at 8.57 ± 0.79% (w/w) compared to control at 9.08 ± 0.78% (w/w). Analysis of the medium conducted as an indication of nutrient bioconversion by patchouli culture showed a decrease in pH, conductivity and sucrose content
