Summary:Abstract: <br /> <br /> <br /> Diosgenin was found in six species of 3.75 years old Costus rhizome, namely Costus speciosus Bogor, Costus speciosus Jawa, Costus speciosus Australia, Costus malortieanus, Costus rumphianus, Costus villisissinus, Costus discolor, Costus speciosus New Guinea and Costus afer obtained from Balai Penelitian Tanaman Industri Bogor. <br /> <br /> <br /> Determination of diosgenin contents in Costus rhizome by thin layer chromatography densitometry indicated 1.41% (Costus speciosus Bogor), 0.66% (Costus speciosus Jawa), 0.85% (Costus speciosus Australia), 0.64% (Costus malortieanus), 0. 83% (Costus rumphianus), 0.90% (Costus villisissinus), 0.64% (Costus discolor), 0.86% (Costus speciosus New Guinea) and 0.76% (Costus afer), and determination by spectrophotometry indicated 3.03% (Costus speciosus Bogor), 1.20% (Costus speciosus Jawa), 1.10% (Costus speciosus Australia), 0.98% (Costus malortieanus), 1.23% (Costus rumphianus), 1.05% (Costus villisissinus), 0.91% (Costus discolor), 1.06% (Costus speciosus New Guinea) and 1.20% (Costus afer) calculated on a dry weight basis. <br /> <br /> <br /> Determination of diosgenin contents in Costus rhizome after incubation with water for twenty four hours at room temperature by thin layer chromatography densitometry indicated 3.13% (Costus speciosus Bogor), 0.67% (Costus speciosus Jawa), 1.54% (Costus speciosus Australia), 0.96% (Costus malortieanus), 1.13% (Costus rumphianus), 0.97% (Costus villisissinus), 0.72% (Costus discolor), 1.43% (Costus speciosus New Guinea) and 1.73% (Costus afer) and determination by spectrophotometry indicated 3.83% (Costus speciosus Bogor), 1.33% (Costus speciosus Jawa), 2.06% (Costus speciosus Australia), 1.15% (Costus malortieanus),1. 32% (Costus rumphianus), 1.41% (Costus villisissinus), 0.96% (Costus discolor), 1.62% (Costus speciosus New Guinea) and 1.29% (Costus afer) calculated on a dry weight basis. <br /> <br /> <br /> Distribution of diosgenin contents in Costus species rhizome was as followed: the outside part of rhizome bordered by endodermis contained 1.41% (densitometry) and 3.03% (spectrophotometry), and the inside part bordered by endodermis contained 1.19% (densitometry) and 1.22% (spectrophotometry). <br /> <br /> <br /> The saponin mixtures obtained from Costus speciosus showed 9 spots on thin layer chromatography using silica gel G, chloroform methanol water (60:35:10) and vanillin sulphate acid spray reagent. The hydrolisis of saponin from nine samples of Costus on thin layer chromatography showed diosgenis and two unidentified compounds. <br />
