Meanwhile, turmeric extract contains curcumin compounds which have anti-inflammatory effects. This study aims to see the anti-inflammatory effects of a combination of ESM and turmeric extract gel emulsion (emulgel) using rat foot edema parameters. ESM was enzymatically hydrolyzed using papain enzymes and sodium sulfite. ESM hydrolysate (ESMH) was fractionated using membrane ultrafiltration. The ESMH <10 kDa fraction and turmeric extract were formulated into an emulgel preparation. Emulgel evaluation consisted of organoleptic, pH, viscosity, and homogeneity. Anti-inflammatory activity tests were carried out using two methods, namely chronic and acute inflammation. In each test, there were five test groups, consisting of negative control (no treatment), positive control (glucosamine-chondroitin cream), test group I (emulgel I, ESMH <10 kDa fraction 0.3%), test group II (emulgel II, ESMH <10 kDa fraction 0.6%), and test group III (emulgel III, ESMH <10 kDa fraction 0.6% + turmeric extract 0.6%). Acute anti-inflammatory tests were performed using 0.05 mL of 1% carrageenan suspension injected into the left hind paw of the rat intraplantarly. Observations were made every 30 minutes for 5 hours. Chronic anti-inflammatory tests were carried out using 0.2 mL of Freund's Complete Adjuvant (FCA) emulsion 1:1 injected into the same area. Edema observations were carried out until the 28th day. In the acute anti-inflammatory test, emulgel II (ESMH fraction <10 kDa 0.6%) and emulgel III (ESMH fraction <10 kDa 0.6% + turmeric extract 0.3%) gave significantly different %edema values to the negative control group (P<0.05) using the AUC parameter starting from time 0 to 240. In the chronic anti-inflammatory test, all emulgel preparations did not give significant changes in %edema to the negative control group (P>0.05). text |
