Forster & J.G. Forster is one of the main genus of Moraceae.. It is generally known as breadfruit trees and comprises about 47 species. This genus is a source of edible fruits, besides that, it is also used for its wood, stem bark and gum. In Indonesia, it is also used as traditional medicines. The role of the plants in curing many diseases has a relation with their secondary metabolites. Artocarpus contains phenolic and non phenolic compounds including flavonoid, stilbenoid, arylbenzofuran and neolignan. Prenylated flavonoids are the major constituents of Artocarpus. Artocarpus scortechinii King is one species of Artocarpus growing in Indonesia. It has been reported that only the stem bark of A. scortechinii which have been examined for its chemical contents. In this research, by using extraction method and purification by various of chromatography techniques, have isolated five compounds from the root bark of A. scortechinii. Those compounds are artonin E, artonin V, cycloartobiloxanthone, artoindonesianin Z-5 and 12-hydroxyartonin E. The structure elucidation of all compounds are based on spectroscopy data including UV, IR, 1H and 13C NMR. Artonin V, 12-hydroxyartonin E and artoindonesianin Z-5 were for the first time isolated from A. scortechinii. However, artonin E and cycloartobiloxanthone have been found previously from the wood bark of the same species. The cytotoxicity toward murine leukemia P-388 cells showed that four compounds, i.e. artonin E, artonin V, artoindonesianin Z-5 and 12-hydroxyartonin E have very strong activity with IC50 0.06 μg/mL, 1.1 μg/mL, 1.8 μg/mL and 1.7 μg/mL, respectively. Artonin E is the most active compound among the isoprenylated flavones. One compound, cycloartobiloxanthone, is not active with IC50 4.6 μg/mL. text
