<br />
From 1880 up to now thousands of people in Banyumas and Kediri died in cases of bongkrec poisoning. <br />
<br />
<br />
The bongkrec bacteria produce poisonous compounds in the medium of tempe bongkrec and especially in the partly defatted coconut, in other words,tempe made from pressed coconut shred can be poisoned by this bacteria. <br />
<br />
<br />
Pseudomonas cocovenenans can be introduced as a contamination before or during fermentation . Under the conditions of the manufacturing process, these bacteria are able to produce toxins from the coconut substrata These include the colourless bongkrec acid, for which the LD 50 for mice after injection is 1,4 mg per kg bodyweightand a toxoflavin ( mp 171 C), for which the LD 50 for mice after injection is 1,7 mg per kg of body weight. <br />

A crude ethanolic extract of pressed coconut shred fermented with Pseudomonas cocovenenans was found to inhibit the respiration of bovine heart homogenat ( p > 0,05 ). <br />

<br />
The aim of this research was the isolation and purification of some compounds with antibiotic activity which are produced by Pseudomonas cocovenenans strain X 128 liophylized in Bogor ( BCC 2184 ). <br />
<br />
<br />
Three toxoflavin have been successfully isolated and purified using the method described by Soedigdo with some improvements, while thin layer chromatography was used for the identification of the three coumpounds. <br />
<br />
<br />
The antibiotic activities towards Escherichia coli and Aspergillus niger were determined with the disk and dilution methods. <br />
<br />
<br />
Three pure compounds were found. The characteristics of the three compounds which have antibiotic activity are : <br />

The factor of retention of the first compound in chloroform : methanol 1 : 1 was about 0,75. It showed white fluorescence and a melting point of 2160 C. The ultra violet spectroscopy showed two maxima at 268 nm and 360 nm. The infra red spectra showed the following peaks ; 2800, 2900, 1530, 1550, 1460, 1120, 720 cm-l. <br />

The second compound showed a factor of retention in chloroform : methanol 1 : 1, of about 0,65, it showed blue - black fluorescence and yellow needle cristals , melting point 205 C. <br />
<br />
<br />
In the ultra violet spectrum showed two maxima at 258 nm and 202 nm. The infra red characteristic are ; 700,740, 850, 880,930, 980, 1020,1080, 1120, 1170, 1220, 1260, 1310, 1320, 1460, 1490,1530,1560, 1600,1680, 1770, 2580, 3050, 3500 cm-l. |
