STUDI PENDAHULUAN EFEK SINAR ULTRA VIOLET U.V.) TERHADAP MUTAGENESIS STREPTOMYCES SP.

Preliminary investigation on the muta€enic effects of Ultra Violet radiation on a Strectomyces 2 strain isolated from Indonesian soil was conducted. A B.V. dose-survival spore curve was constructed, showing a corelation at F = 0,001 and 4.x = 7 of the number of survival spores to the dosage of L.V....

By irradiating spores with U.V. light giving a high kill of 90 - 98%, one major marfological mutant was obtained from 302 survival spores examined. This mutant designed as Nor-1, was stable for the period since isolation till the end of the studi. At this same dosage, no mutants resistant to 100 mikrograms of Streptomycin per ml media was obtained from approximately 640 survival spores. Media M5D-R was used as the minimum media, andhidrolitik action towaras amylum w i l d protein and the lipolitik action was observed. tor-1 and Mor-4 indicated an increased ability to hidrolise amylum as to that shown by the wild type ael which showed a poor ability to hidrolise amylum. ;While Mor-1, Nor-3, Nor-4 and the wild type cell all showed lip olitic action, non was observed from u.._-_.
