Summary:Abstract: <br /> <br /> <br /> <br /> <br /> Cellulolytic enzymes are the enzymes which catalyze the hydrolysis of 0-1,4 glycosidic bonds of the cellulose and its derivatives. There are many different cellulolytic enzymes such as avicelase, carboxymethyl cellulase (CMCase) and cellulase, which are produced by cellulolytic microbes living freely in nature or in animals intestines. It has been reported that cellulose-degrading microbes are able to degrade the cellulose derivatives but not necessarily vice versa. Isolating cellulolytic microbes usually uses cellulose as a carbon source, instead of cellulose derivatives, yet there is no explanation on which cellulose derivative cannot be used. <br /> <br /> <br /> <br /> <br /> According to the literature, cellulolytic enzymes produced by one type of microbe is coded by different genes, in both the amount base pairs and the nucleotide sequences. Furthermore, the genetic expression of cellulolytic coding genes was influenced by the availability of cellulosic substance in the growth media. However, there is no explanation available on the interaction of the cellulolytic coding genes during the enzyme biosynthesis process. <br /> <br /> <br /> <br /> <br /> If the degrading microbes of cellulose derivatives can degrade cellulose and cellulose degrading-microbe can degrade cellulose derivatives, even more cellulolytic enzymes can be produced simultaneously within the same enzyme preparate, it will be indicate that there is an interaction among the cellulolytic enzyme coding-genes during the process of enzyme biosynthesis. This interaction should be investigated to obtain some explanation on how the cellulolytic enzyme biosynthesis is regulated. <br /> <br /> <br /> <br /> <br /> The purpose of this research is to study how cellulolytic enzymes are produced by a cellulolytic microbe. The study involves some experiments as follows : (1). Isolating cellulolytic microbes using CMC as the only carbon source The optimum fermentation condition of P. alcaligenes PaAf-18 to produced cellulolytic enzyme was determined by conducting several fermentations at various temperatures, pH and incubation period. The resulted enzymes were then isolated and cellulolytic enzyme activity tested. The result showed that this bacteria produced cellulolytic enzymes optimally at 37 farenheit C, pH 7,0 with the period of 7 - day- fermentation.
