From about 28 species of Stephania which has been investigated for their alkaloid content, it is apparent that most of the alkaloids could be theoritically derivable from benzylisoquinoline as the precursor. The present investigation concerns with the isolation of alkaloids from the stem of the plant Stephania corymbosa 81, which is used traditionally to cure fever. The isolation was carried out based on the generally basic character of alkaloids, and the process involved several steps, namely removal of lipid from the plant, isolation of alkaloid, separation of phenolic and non-phenolic alkaloidal constituens, and separation of constituents by column chromatography methods. The purity of the isolated compounds were determined by thin layer chromatography ultilizing specific reagent for alkaloid detections. Ultra violet, infra red, proton nuclear magnetic resonance and mass spectroscopy data indicated that one of the isolated alkaloid is a dibenzazonine alkaloid. It is also concluded that the alkaloid is protostephanine . This work is the second occassion when protostephanine was isolated from the genus Stephania, the first was from Stephania japonica Miers, discovered by Takeda in 1950. Protostephanine has also been isolated recenth from another spesies of Hyperbaena columbicao of Menispermaceae family, however it has never been isolated from other plants. text |
