The purpose of this research is to analyze the amount of a-tocopherol that is one of the antioxidant agent in rice bran oil. The a-tocopherol is identified an quantified by High Performance Liquid Cromatography (HPLC) method with optimum condition as followed: mobile phase of methanol : asetonitril (4:1); isocratic elusion; Octadecyl Silane (ODS) column; flow rate 0.8 ml/minutes; spectrophotometric detector at wave length of 292nm and a-tocopherol was eluted in 5.10 minutes. Linearity test of calibration curve yielded a validation data of correlation coefficient (r) = 0.999 and variation coefficient of line regression (V0x) = 1.69%. Percent recovered test result was 96.87% at concentration level of 6.97 ug/ml. The method gave Relative Standar Deviation (RSD) respectively 1.94% at concentration level of 10 ug/ml with limit of detection and limit of quantization of 2.53 and 7.6 ug/ml. The A-tocopherol amount in rice bran oil is 1.18+0.01 mg/g. <br /> text |
