Summary:Research on the inhibitory activity of free and non-freee alkaloids 70 % ethanolic extract of leaves of Justicia gendarussa Burm.f against HIV reverse transciptase enzyme has been reported. The aim of the research was to find a solution to face the high prevalency of HIV AIDS with its side effects, resistances problem and the mechanism of inhibition of HIV Reverse Transcriptase enzyme. Justicia gendarussa has already known to having an antiHIV effect, and that’s why we are also looking the inhibition mechanism. Both types of extracts tested in vitro using Elisa technique and analyzed chemical content of Gendarusin A as antiHIV using high performance liquid chromatography. Elisa test results obtained percent inhibition, respectively 48.23, 35.10, 35.35, 27.78 and 26.01 for the concentration of 250 ppm, 125 ppm, 62.5 ppm, 31.25 ppm and 15.63 ppm of non alkaloids-free extract and 48.74, 48.48, 40.40, 32.83 and 30.56 for the concentration of 250 ppm, 125 ppm, 62.5 ppm, 31.25 ppm and 15.63 ppm are alkaloid-free extract. The IC50 of non alkaloids-free extract and alkaloid-free extract respectively are 393.02 ppm and 220,98 ppm. Both type of extract inhibit activity of HIV Reverse Transcriptase uncompetitively. The results high performance liquid chromatography obtained Gendarusin A in the non alkaloid-free extract are 3.54 % and alkaloid-free extract are 2.30 %. Therefore, these results conclude that the Justicia gendarussa Burm.f can be a useful source for the isolation and development of new inhibitors antiHIV.
