Summary:Klebsiella pneumoniae is an important bacterium that most commonly transmits antibiotic resistance genes. The blaTEM gene is the most frequently detected gene in K.pneumoniae, which is most widely distributed in food-producing animals. In several studies that have been conducted, blaTEM is the dominant gene found from K.pneumoniae isolates which from pigs. This study aims to detect the presence of the blaTEM gene in K.pneumoniae isolated from pigs rectal swabs. Each sample was cultured on Mac Conkey Agar media and identified by morphological and biochemical tests. K.pneumoniae positive was examined with antibiotic sensitivity testing using the Kirby-Bauer method against the antibiotics ciprofloxacin, streptomycin, trimethoprim, tetracyclin and aztreonam. The PCR test was continued to detect the presence of the blaTEM gene as the ESBL encoder. The results obtained in this study were 8 of 130 samples of pigs rectal swabs (6.2%) tested positive for K.pneumoniae. For antibiotic testing, it was found that K.pneumoniae was resistant to tetracyclin and trimethoprim antibiotics 87.5% (7/8), streptomycin antibiotics 75% (6/8) of intermediate isolates and 25% (2/8) of isolates were resistant. All K.pneumoniae positive isolates were still sensitive to the antibiotics ciprofloxacin and aztreonam. It was found that 1 out of 8 isolates of K.pneumoniae MDR (12.5%) were positive for the ESBL coding blaTEM gene. This study has succeeded in isolating K.pneumoniae bacteria from swab rectal in pig farms in Gresik and Malang Distirics and succeeded in confirming the presence of K.pneumoniae isolates MDR, accompanied by the findings of K.pneumoniae that were positive for the blaTEM gene as an ESBL encoding for the MDR K.pneumoniae isolate.
