Summary:Background: Noni fruit (Morinda citrifolia L.) has long been known by the people of Indonesia, especially Java, Sumatra and Borneo. Noni fruit (Morinda citrifolia L.) is also widely used as traditional medicine in almost all islands in Indonesia. Noni fruit (Morinda citrifolia L.) is also known to have antibacterial activity against Staphylococcus aureus and MRSA (Methicillin-Resistant Staphylococcus aureus). The anti-bacterial activity of noni fruit is supported due to the identification of phytochemical compounds such as iridoids, flavonoids, fatty acids, lignans, anthraquinone vitamins and trace elements. Objective: The purpose of this study was to determine the antibacterial activity methanol extract of Noni fruit (Morinda citrifolia L.) against Staphylococcus aureus and MRSA (Methicillin-Resistant Staphylococcus aureus). Methods: This research was an experimental laboratory using a dilution and well-diffusion method to determine the antibacterial activity methanol extract of Noni fruit (Morinda citrifolia L.) against Staphylococcus aureus and MRSA (Methicillin-Resistant Staphylococcus aureus) in vitro. For the dilution method there are nine concetration from 100%; 50%; 25%; 12,5%; 6,25%; 3,125%; 1,5625%; and 0,78125%. Minimum Inhibitory Concentration (MIC) values and Minimum Bactericidal Concentration (MBC) values were determined by dilution method. The MBC values were determined by suspension streaking from muller hinton broth on agar plate. There are five concentrations were used for the diffusion method, which were obtained from serial dilution using distilled water. The concentrations used were 100%, 50%, 25%, 12.5%, and 6.25%, respectively. The positive control of this study for Staphylococcus aureus was erythromycin 15 μg, and for MRSA was vancomycin 30 μg. Results: Minimum Inhibitory Concentration (MIC) of methanol extract from Noni fruit (Morinda citrifolia L.) on Staphylococcus aureus and MRSA (Methicillin-Resistant Staphylococcus aureus) in this study could not be determined due to the extract were dark green in color. Meanwhile, the Minimum Bactericidal Concentration (MBC) of methanol extract from Noni fruit (Morinda citrifolia L.) on Staphylococcus aureus and MRSA (Methicillin-Resistant Staphylococcus aureus) in this study was 6,25% and 3,13% respectively. From this research it shows a higher antibacterial activity againts MRSA. The average range diameter of the inhibition zone in each spesific case was 0 – 26.64 mm for Staphylococcus aureus and 0 – 24.63 mm for MRSA. Noni fruit extract had a higher inhibition zone ratio against MRSA when compared to the positive control antibiotics for each bacteria. Noni fruit extract showed higher antibacterial activity against MRSA compared to Staphylococcus aureus when viewed from MBC and comparing the largest inhibition zone with positive controls. Therefore, further research is needed in the development of Noni fruit (Morinda citrifolia L.) as an oral antibiotic or topical antibiotic.
