Summary:The purpose of this reasearch was to determine the effect of vitamin E (α-tocopherol) on the number of Sertoli cells after being exposed to 2,3,7,8-Tetrachlorodibenzo-p-dioxin (TCDD) in white rats (Rattus norvegicus). This is an experimental research with completely randomized design. Twenty five male white rats strain Wistar with average weight around 200 gram were used in this research. White rats were divided into 5 group of treatment (5 rat each group), consisting of one control group K(-) and four treatment groups P0, P1, P2, and P3, treated with different combination of TCDD and vitamin E (α-tocopherol) administered through oral gavage for 21 days. Histopathology slides of testis were made with HE staining and continue with the calculation of Sertoli cell. The data were analyzed using One Way ANOVA and continue with Duncan test (p< 0,05). The result of the research showed that K(-) group had the highest number of Sertoli cells, while P0 group had the least number of Sertoli cells, P3 group which given vitamin E (α-tocopherol) 259 mg/kgBW/day is the most effective method in maintaining the number of Sertoli cells, because P3 didn’t show a significant difference (p>0,05) with the K(-) group which was only given 0,5 ml/day of corn oil. Giving vitamin E (α-tocopherol) to white rats after being exposed to TCDD can maintain the number of Sertoli cells, because vitamin E (α-tocopherol) has antioxidant efficacy in reducing TCDD-induced AhR activation and they can prevent the production of cell membrane lipid peroxidation. Thus, TCDD can be used to prevent and treat acute and chronic TCDD intoxication in the male reproductive organs.
