Summary:"Objective : Tumor growth and metastasis in NPC patients suspected role of several molecular biomarkers that have been identified in tumor specimens of patients with NPC. Recent research states p16 gene inactivated cause of mutation can be a indicator of prognosis and able to prepare better therapy for NPC. The purpose of this study is to analyze inutation of pl 6 gene of NPC patients which associate with histopathological type ofNPC. Methods : Biopsy specimens divided into two part, first part for pathology examination and second part for PCR. Histopathological type were obtained from 21 NPCs with Meyer,s hematoxillin eosin staining divided into three type, WHO type 1, type 2 and type 3. Assesment of histopathological type was perfomed by pathologist consultant. The mutation of p16 gene was obtained with polymerase chain reaction from primary tumor tissue with termal cycler machine Bioneer and sequencing with ABI PRISM 310 machine. Assessment of mutation was performed by consultant staff from Institute of Tropical Disease Airlangga university Surabaya. The Spearman's test was used to determine the relationship between mutation of p16 gene and histopatological type of NPC. Statistical significance was defined as p < 0.05. Result : There were 21 NPC patients that met the inclusion and exclusion criteria. Showed negative mutation of p16 gene by 19 (90.48%) for all samples, and 2 (9.52%) with positive mutation of p16 gene which all mutation were WHO type 3. Spearman test results scores p= 0.568 with a correlation coefficient -0.132. Association mutation of p16 gene with histopathological type (WHO type 1, 2, 3) in patients with NPC showed not significant > 0.05). Conclusion : There wa.s not association between mutation of pl 6 gene and histopathological type of nasopharyngeal carcinoma. "
