Summary:The purpose these research to prove the alkaloid Acyranthes aspera linn to influence on viability, apoptotic inductio, necrotin induction, lethal concentration 50% (LC50) and optical density of caspase by Elisa on myeloma cell. In this research was found 97,73% alkaloid fraction of Achyranthes aspera linn by high performance liquid chromatography. Alkaloid fraction of Achyranthes aspera linn was prepare by Pharmacope Indonesian methode. The effect of alkaloid was proved on myeloma cell, devided into 3 groups: the first group was treatment groups, was given alkaloid fraction in concentration 1 ppm, 10 ppm, 100 ppm and 1000 ppm. The second group was negative control was given RPMI and OMSO media only. The third groups was positive control was given colchicin in concentration 100 ppm. Each groups was represented with 4 hole in 24 microwell plate hole.The result showed that the alkaloid fraction of Achyranthes aspera linn in dose 100 ppm in vitro cause viability of myeloma cell 9,45 ~ 5,96% lower than positive control 13,18 ± 3,67%, caused apoptotic 37,52 ~ 3,67% and higher than positive control 31,41 ~ 5,96%, caused necrotic myeloma cell 53,03 ~ 2,42% lower than positive control 55,40 ~ 4,23% and the lethal concentration 50% on myeloma cell was 0,719 ppm. The optical density of caspase by Elisa test to begin at dose 10 ppm concentration of alkaloid Achyranthes aspera linn (value of caspase > 2x COV). The conclution of these research the valkaloid Achyranthes aspera linn to cause of cell death of myeloma. Mechanism of action by necrosis and apoptotic, and increase of dose of alkaloid to cause of increase the optical density of caspase.
