Summary:Purposes : To evaluate the effectiveness and the mechanism of Swim up (SU method) and Swim up + Aside migration (SU + ASM) combination method on spermatozoa quality (motility, live spermatozoa and normal morphology) and spermatozoa fertilizing ability (in vitro fertilization rate and embryo cleavage rate), also an increasing of V-chromosome bearing spermatozoa ratio of Bali Cattle based on spermatology paradigm with the of time, motility and in vitro fertilization. Methods : The type of this research was laboratoric true experimental SU method (30, 45 and 60 minutes) and SU + ASM combination ( 30 minutes SU and 30 minutes ASM) on a number of samples devided into three groups. These groups consisted were one control group (K) and two treatment group (group -I and -II). Independent variable in this research were SU (30, 45 and 60 minutes) and SU + ASM combination (30 minutes SU with 30 minutes ASM). Dependent variable comprised to (a) sperm quality (motility, live spermatozoa and normal morphology); (b) in vitro fertilization, (c) in vitro embryo cleavage rate and (d) Y - chromosome bearing spermatozoa ratio based on head size and chromosomal analysis. Intervening variable was the oocyte maturation. Results : The result showed a significant difference by using Manova (p~O,05) in sperm motility, live spermatozoa and normal morphology established the highest normal sperm morphology, compared with the other groups. The result of Anova test revealed a significant difference (p~O,05) in fertilization rate between treatment group and control group. SU 30 and SU 45 treatment group was significantly higher, when compared to SU 60 and SU+ASM combination groups. From Anova analysis, it was found that there was a significant difference (p~0,05) in with control, SU 45 and SU 60 groups, at the level of embryo development observed 48 hours after fertilization. it was showed that in all of the treatment groups, embryos developed ~ 4 cells were significantly higher (p~0,05) than embryos developed> 4 cells, whereas no any difference was found between quantity of blastomer cells in the control group. According to Anova analysis, it was showed that Y -chromosome bearing spermatozoa in treatment group were significantly different (p~0,05), compared with control groups.
