Summary:It is well-known that acute eccentric exercise / downhill running causes significant muscle damage in healthy mice (Mus musculus) but its effect in decreasing GLUT-4 protein in muscle is still debatable. The purpose of this study was to prove the effect of eccentric exercise / downhill running at slope angles of -5 ˚, -10 ˚ and -15 ˚ in relation to decreased GLUT-4 protein level in gastrocnemius muscle of healthy mice.This study was a laboratory experimental research, using the Randomize Post Test Only Control Group Design method. We used strains balb / c adult mice (Mus musculus ), male, with 8-12 weeks of age, healthy and active, body weight between 20-25 grams. The total amount of 28 trials animals were randomly divided into 4 groups. The control group ( K0 ) ran on a treadmill at slope angle of 0 ˚, the downhill running group at slope angle of -5 ˚ ( K1 ), the downhill running group at slope angle of -10 ˚ ( K2 ) , and the downhill running group at slope angle of -15 ˚ ( K3 ) . Exercises performed using a treadmill for 18 minutes including warming up for 5 minutes at a speed 31cm/second. Body weight and GLUT � 4 were taken from each trial group. In this research, measurement of decreased GLUT � 4 levels in mice gastrocnemius muscle using the ELISA method. Muscle GLUT - 4 levels ( nm / mg ) was obtained from the absorbance value of GLUT - 4 ( nm / ml ) divided by the total muscle protein content ( mg / ml ). The mean of GLUT-4 levels in K0 (0.329 ± 0.162) lower than that of all treatment (downhill running) groups; K1 (0.959 ± 0.557), K2 (1.020 ± 0.359) and K3 (0.943 ± 0.441). The result showed a significant difference between the control group and all of three treatment (downhill running) groups; K0-K1 (p = 0.008), K0-K2 (p = 0.004) and K0-K3 (p = 0.009) while GLUT-4 levels in all of three treatment groups showed no significant differences; K1-K2 (p = 0.779), K1-K3 (p = 0.943) and K2-K3 (p = 0.725). The conclusion of this research is acute eccentric exercise / downhill running at slope angles of -5 ˚, -10 ˚ and -15 ˚ increasing muscle GLUT-4 levels in healthy mice, but there were no any different between all of slope angle group.
