ISOLASI DAN KARAKTERISASI ENSIM SELULASE DARI KEONG EMAS DAN RAYAP SEBAGAI BAHAN PENDEGRADASI SELULOSA

The aim of this research is to know the type of celullase enzyme from golden snail's gut and termite's gut and to know the enzyme activity by in-vitro system. Supernatant from golden snail's (Pomacea canaliculata) gut and termite's (Macrotermes sp.) gut were characterized the en...

ANAM AL-ARIF,, 131836993 SF600-1100 Veterinary medicine The aim of this research is to know the type of celullase enzyme from golden snail's gut and termite's gut and to know the enzyme activity by in-vitro system. Supernatant from golden snail's (Pomacea canaliculata) gut and termite's (Macrotermes sp.) gut were characterized the enzymes type by SDS-PAGE, Western Blot and Dot Blot testing. The enzymes activity from golden snail and termite's supernatant then tested by CMC, pNPC and pNPG. The result of this research were there are many protein bands on SDS-PAGE that estimate of cellulase enzyme type, that is exoglucanase, endoglucanase and fl¬glucocidase enzymes. Western Blot and Dot Blot test then used to ensure this estimation. Western Blot test didn't clear showed the protein bands, while Dot Blot test showed the antigen-antibody reaction from golden snail and termite cellulase same as purified cellulase from Trichoderma virile, that revealed exoglucanase, endoglucanase and 13-glucocidase enzymes. The golden snail's endoglucanase activity was higher than termite's endoglucanase activity. The golden snail's exoglucanase activity was higher than termite's exoglucanase activity. The golden snail's [3-glucocidase activity was higher than termite's 3-glucocidase activity. The conclusion of this research were there are 3 cellulase enzymes type in golden snail and termite gut, that is exoglucanase, endoglucanase and ll-glucocidase enzymes. The golden snail's cellulase enzymes activity was higher than termite's cellulase enzymes activity.
