Summary: | Abstract Cumulus cells and liver cells (hepatocyte) have an ability to produce IGF-1 through monolayer culture, but the concentration and the bioactivity in embryo development was not clear. The aim of this study was to analysis and bioactivity test of IGF-1 there found in product cell. Liver cells culture of cattle was prepared by grinding liver tissue followed by trypsination and repeatedly centrifuged were obtained hepatocyte. Cumulus cells were obtained from aspirating ovarian follicles with diameter 2-6 mm. Liver and cumulus cells were cultured in TCM 199 + FCS 10% + BSA 10%. Cell concentration was 1.9 x 106 /ml medium then culture was incubated in incubator at 38.5oC 5% CO2 for 3, 6, 9 and 12 days. IGF-1 concentration after culture was measured by IRMA. The result of research showed that 6 days incubation period resulted the most confluent monolayer compared to the other incubation period, either in liver or cumulus cells, therefore resulting the highest concentration of IGF-1. In liver cells IGF-1 concentration was higher compared to these of the cumulus cells, meanwhile in cumulus cells the estrogen was found higher than IGF-1. The conclusion of this research was that monolayer culture of bovine liver and cumulus cells could produce IGF-1. Abstrak Sel kumulus dan sel-sel hepar (hepatosit) memiliki kemampuan untuk memproduksi IGF - 1 melalui kultur monolayer, tetapi konsentrasi dan bioaktivitas dalam perkembangan embrio tidak jelas. Tujuan dari penelitian ini adalah untuk menganalisa dan menguji bioaktivitas IGF-1 yang dihasilkan dalam sel tersebut. Sel hepar dan sel kumulus dibiakkan dalam TCM 199 + FCS 10 %. Konsentrasi sel adalah 1,9 x 106 / ml dalam medium kultur diinkubasi dalam inkubator pada temperatur 38.5oC 5 % CO2 selama 3, 6, 9, dan 12 hari. Konsentrasi IGF - 1 setelah dibiakkan diukur dengan IRMA. Hasil penelitian ini menunjukkan bahwa inkubasi 6 hari dihasilkan kultur monolayer paling konfluen dibandingkan dengan masa inkubasi yang lain; baik pada sel hepar maupun pada sel cumulus, sehingga menghasilkan konsentrasi tertinggi IGF-1. Dalam sel-sel hepar konsentrasi IGF-1 lebih tinggi dibandingkan dalam sel cumulus; sedangkan pada sel-sel kumulus estrogen ditemukan lebih tinggi dari IGF-1. Kesimpulan dari penelitian ini menyatakan bahwa kultur monolayer sel hepar dan sel kumulus sapi dapat menghasilkan IGF-1
