Summary:METHOD VALIDATION OF GAS CHROMATOGRAPHY WITH FLAME IONIZATION DETECTOR FOR DETERMINATION OF ETANOL IN LEGEN OR SIWALAN (Borassus flabellifer Linn.)SAP AdhadiAM Siwalan (Borassus flabellifer Linn.) sap or legen is one of the Indonesian traditional beverage.Siwalan sap during storage contains alcohol (ethanol) as the product of fermentation by enzimatic or microorganisms. The amount of alcohol in alcoholic beverages can affects central nervous system (CNS). The purpose of this research is obtaining validated method of gas chromatography with flame ionization detector for determination of etanol in siwalan sap. Gas chromatography Agilent 6890 Series GC system, with a Flame ionzation Detector (FID) and a coloum HP-5 was used in this study. Optimal condition were obtained by using temperature inlet and detector of 250°C, whilst the temperature of oven programed at 55 – 120°C. Split ratio was set at 50:1 and the injection volume was 1.0 μl.The method was proved to be selective. Linearity showed linear respone (r = 0.9999) while correlation coeffecient of the function (Vxo)was 2.4%over the range of concentration used (between 0.025% to 2%). Accuracy of method was in range of accepted limits (90– 108%), recovery was found to be in the average of 97.12%, while the precision was 1.51%. The detection limit is 5.68 x 10-4%, and the quantification limit is 1.72 x 10-3%. The proposed method was successfully applied for the analysis of ethanol in siwalan sap.Siwalan sap was stored until five days in room temperature, extracted by appropriate solvent, and analyzed with gas chromatography method.All of them showed the profile of alcohol concentration. The result of quantitative analysis of ethanol content in siwalan sap in five different days of storage were 0.5694, 2.429, 4.357, 5.678, 5.894 and 6.292%.
