Description
Summary:The purpose of this research was to know the characteristic of Bacillus sp. A32c bacteria based on morphology test, biochemical, and growth pattern. The method which was used on this research was descriptive method, which was doing the data analysis and presenting in growth curve shape of the bacteria. The result of this research showed that Bacillus sp. A32c had yellow ish white colony, the surface of the colony was having no mucus, and the shape of the colony was circle with emerged edge, the edge of the colony was wavy with emerged elevation angle. The cell morphology test showed that Bacillus sp. A32c bacteria was Gram positive bacteria with single bacil shaped cell, having endospore with ellipse shape which was in center cell, and was motile bacteria. Biochemical test showed that Bacillus sp. A32c on amylum hydrolysis test was positive, VP test was negative, and citrate test was negative. On the observation of growth pattern, the growth pattern of Bacillus sp. A32c showed three phases which are lag phase, log phase, and stationary phase. The lag phase started at the 3rd hour until 12th hour, the log phase started at 12th hour until 39th hour, meanwhile stationary phase started at 39th hour. The dead phase could not be observed because the dead bacteria cell would be still counted the optical density value. Because of that, to see the perfect growth pattern, further research is needed by using Total Plate Count (TPC) method so the dead phase can be observed.
