Summary:This study aims to determine the growth pattern of Bacillus sp. C32f in Mineral Salt Medium, the beginning of the stationary phase is a phase that produces secondary metabolites, and identified Bacillus sp. C32f with morphological and biochemical tests. Bacillus sp. C32f rejuvenated on the Nutrient Broth medium then cultivated on the Nutrient Agar medium. Inoculation of Mineral Salt Medium was performed after isolation of the isolates. The growth pattern of Bacillus sp. C32f is seen based on the growth curve. Method of measuring growth curve using spectrophotometer with wavelength 580nm. Fermentation broth making technique every 3 hours as much as 3ml in duplo for 48 hours. The growth curve shows that bacteria are able to grow on Mineral Salt Medium and have growth patterns consisting of lag phase, exponential phase, stationary phase and death phase. The lag phase occurs at 0-12 hours, and the exponential phase at 15 hours. Initial stationary phase which is the phase where secondary metabolites are produced at 27th hours. Death phase is not visible due to weakness spectrophotometer is a live and dead bacterium is calculated. Identification of Bacillus sp. C32f in Nutrient Agar medium by looking at the characteristics of colonic morphology, cell (Gram stain and spore), and motility on SIM medium and physiology characteristics by means of biochemical test based on Bergey’s Manual of Determinative Bacteriology flowchart include starch hydrolysis test, Catalase test, VP test, Citrate test and Sugar Fermentation test. The results show that Bacillus sp. C32f is suspected to be a link between Bacillus insolitus and Bacillus marinus.
