SEBAGAI AGEN ANTIKANKER PADA KANKER KOLON (WiDr Cell Line) PENELITIAN EKSPERIMENTAL LABORATORIS AMAQ FADHOLLY, 061714453003 SF600-1100 Veterinary medicine This purpose of this research was to manufacture nanoparticles of Annona squamosa leaf extract which containing alcaloids, flavonoids, saponins that have cytotoxicity of WiDr cell lines through in vitro test and continued with IHC assay to know the expression of caspase-3 protein as pro-apoptosis protein and double staining assay to know the apoptosis activity. Nanoparticles was made by ionic gelation method and tested with PSA and FITR. Treatment nanoparticles of Annona squamosa leaf extract was administered to WiDr cell lines at doses of 25 μg/ml, 50 μg/ml, 100 μg/ml, 200 μg/ml and 400 μg/ml. The results showed that nanoparticles of Annona squamosa leaf extract has cytotoxicity in WiDr cell lines with IC50 197,23 μg/ml and it collerated with IHC assay that showed the expression of caspase-3 protein increased followed the doses. Apoptotic activity increased and it was showed by the expression of cells have orange colours.
