Summary: | Background: Amylase production has been investigated using bacteria isolated from soil Api Kayangan Bojonegoro, East Java. Objectives: Objective: This study aimed to isolate and identify the bacteria and investigate the effects of culture conditions on amylase activity. Methods: The bacteria were cultured in media containing soluble starch as carbon source. The addition of calcium (15 mM) or yeast extract (0.5%) and peptone (2%) to the media and the starch dissolved minerals will reduce the time lag phase and extend the growth and production of amylase. Glucose added to the culture reducing the production of amylase, so that the effect of glucose on these organisms. Results: Initial pH media and optimum temperature on amylase production by organisms respectively 7.0 and 50°C. The optimal temperature and pH for the activity of each 50°C and 6.0. Enzyme solution retained 100% activity when incubated at 90°C for one hour and 40% at 60°C for 24 hours. Conlclusion: Glucose in the culture will decrease the production of amylase. |
