This research was aimed to know the effect of tablet manufacturing technique using a combination of polyethylene oxide WSR 1105 (PEO), sodium carboxymethylcellulose and carbopol 940 as mucoadhesive polymer tablet with the ketoprofen as the active ingredient to the quality and physical properties of tablets, drug release profile and discover the optimum formula. Determination of the optimum formula determined by the simplex lattice design, then obtained seven formulas, each formula consist of various concentrations of polyethylene oxide WSR 1105, Carboxymethylcelluloce sodium and carbopol 940. Manufacture of tablets make used of dry granulation technique, and the quality test of granules, tablets, mucoadhesive strength and drug release from the polymers. Data obtained were analyzed by comparing the literature for the requirements of sustained release of drugs, and data analysis was using design expert version 7. 1. 5 to get the optimum formula. Parameters of the optimum formula was obtained compares with the theoretical values by applying the software R 2. 12.2 modul R commander (Rcmdr). Tablet mucoadhesive of ketoprofen was contained combination of PEO, Carboxymethylcelluloce sodium and carbopol 940 polymer extend on characteristic of tablet hardness was in range of between 6,37 - 7,31 kg/cm2, friability within 0,16 to 0,38%, thickness at range on 4,83- 4,92 mm. Uniformity of weight was in the range of 398,3 - 401,0 mg, uniformity of content was in the range of 96,53 to 100,63%. The result showed that the process of dry granulation technique reliable and reproducible. The highest drug release was showed in rasio of concentration carboxymethylcelluloce sodium and carbopol 940 was equal (57,50%) and the smallest released on concentration rasio of carbopol 940 the highest (32,63%). Drug release that followed zero order and first order kinetics. The mechanism of drug release all of formula was followed non-Fickian mechanism (anomalous). Statistical analysis was compared between theoretical and experimental parameters of physical properties give differ significantly.
