Summary:There is no scientific justification about the contents and the benefit of Vanda sanderiana [Rchb.f] Schlechter � Vanda tricolor Lindl. Therefore, this research was aimed to identify potential compounds as anti breast cancer cell lines T47D in Vanda sanderiana [Rchb.f] Schlechter � Vanda tricolor Lindl. vegetative organs. Vanda sanderiana [Rchb.f] Schlechter � Vanda tricolor Lindl. were collected from Vanditia nursery, Sleman. Vegetative organs that used were roots, stems, and leaves and then macerated with chloroform, methanol, and water. Extract toxicities were analyzed using MTT Assay method. Potential extract were fractionated using VLC method. The most potent fraction was isolated using preparative TLC. The most potent PTLC fraction was visualised under UV 254 nm and 366 nm and identified using spray reagents such as cerium (IV) sulphate, liebeman-burchard, vanillin sulphate, dragendorff, and ammonium. The determination of IC50 was calculated using linear regression equation. Signification tests used were one way anova and DMRT (Duncan�s Multiple Rate Test). The results showed that chloroform extract of stem Vanda sanderiana [Rchb.f] Schlechter � Vanda tricolor Lindl. has the lowest IC50 (130 μg/ml) compared with another extracts. The most potent extract was partionated and merged to 8 fraction that tested using MTT assay method to analyzed its cytotoxic activities. E fraction that has the lowest IC50 (55.42 μg/ml) was isolated. The most potent PTLC fraction is fraction middle-low with IC50 value (28.72 μg/ml) lower than doxorubicin (59.85 μg/ml). Potential compounds in fraction middle-low were identified as alkaloids and terpenoids.
