Summary:Daun dewa [Gynura pseudochina (L.) DC.] (Asteraceae) is a native plant of India, Thailand and China and cultivated in Java. The leaves and tubers used as a remedy especially for cancer. Toxicity test against Artemia salina Leach. larvae showed that diethylether fraction of methanolic extract and n-hexane subfraction of diethylether fraction of methanolic extract of G. pseudochina leaves were toxic (LC50 < 1000 µg/mL). According to the problems, evaluation of cytotoxic activity of methanolic extract of G. pseudochina was carried out to explore potential compounds have cytotoxic activity. Cytotoxicity monitoring (Bioactivity-guided isolation) were investigated on Hela and mieloma (NS1) mice cells, using MTT assay. Sohxlet extraction was carried out with methanol solvent, followed by fractionation with chloroform and ethyl acetate in separatory funnel, to give chloroform and ethyl acetate fractions consecutively. The next fractionation were executed by Dry vacuum column chromatography, open column chromatography and preparative TLC, consecutively. Leaves methanolic extract more cytotoxic compare to methanolic extract of its tubers. Continuing investigation with GC- MS analysis of leaves extract and fractions showed that fractions rich in palmitic acid have the highest cytotoxicity against Hela cell with range of IC50 between 101 � 250 µg/mL. Palmitic acid has cytotoxic activity with IC50 = 146 μg/mL and induced apoptosis. This is a new information about Palmitic acid as cytotoxic compound in G. pseudochina. According to structure elucidation by melting point, TLC, spectra data of IR, LC-MS, LC-ESI-ToF MS, 1H dan 13 CNMR, DEPT, two dimention NMR, 1H-1H-COSY, HMBC dan HMQC experiment, from tubers of G.pseudochina have isolated two Alkaloids pyrro- lyzidine, senecionine and senkirkine. The two compounds have weak cytotoxic activity (IC50 > 2000 µg/mL) against NS1 and Hela cells. This is a new information about existence of the two compounds in tubers of G. pseudochina. Based on theoritically approach, senecionine and senkirkine were compounds with hepatoxic activity.
