One of the plants that is known as antioxidant is Rosella (Hibiscus sabdariffa L.). This study aimed to isolate and identify free radical scavenger compound from roselle leaves. Rosella leaves dry powder was maserated with metanol. The dried methanolic extract was then partitioned with n-hexane, CHCl3, ethyl acetate and water and tested for their free radical scavenging activity using DPPH and UVVis spectrophotometer. The active extract was fractionated using vacuum liquid chromatography yielding. Each fraction obtained free-radical scavenging activity was tested and seen TLC profiles. The active compounds in the active fraction isolated using preparative thin layer chromatography. Active compound (isolate 5) was obtained by TLC purity tested with three different mobile phase systems and structure identification based UV and IR spectra data also reagents sliding. Free radical scavenging compounds predicted a flavon with 3, 5, 7, 3�, 4� penta OH and group prenil at 6. This compound is able to inhibit the DPPH radical with IC50 value of 95,01 ± 0,23 μg / ml.
