Summary:Hypertension is the most common serious chronic health problem in adults and carries a high risk factor for arteriosclerosis, strokes, and end-stage renal disease. Hypertensive patients can control their blood pressure by taking medicine to inhibit angiotensin converting enzyme (ACE). ACE plays an important role in regulation of blood pressure in the renin-angiotensin system. Protein of velvet antler of Sambar deer (Cervus unicolor brookei) contains 14 amino acids expected as a source of bioactive peptides that can be used as antihypertensive agent. The stage of this research were : preparation of velvet antler protein, digestion of velvet antler protein by pepsin and trypsin, determination of ACE inhibitory activity, and purification of ACE inhibitory peptides. Digestion process monitoring used some parameters, that were : concentration of soluble protein, peptide content, and profile of molecular weight of native protein fraction as well as after pepsin and trypsin digestion. The result showed concentration of soluble protein (mg/ml) using biuret test was 19.05 ± 0.62 while after pepsin and trypsin were 22.22 ± 0.89 and 16.03 ± 0.84 respectively. Peptide content (mg/ml) of velvet antler was 2.040 ± 0.004 while after pepsin and trypsin were 2.080 ± 0.006 and 2.160 ± 0.003 respectively.
