Description
Summary:Seed health is one of the main factors on long beans (Vigna sinensis L.) cultivation, so seed quality should have important role on the production. BCMV (Bean common mosaic virus) is an important disease that transmitted by seed. Besides BCMV on the long bean, nowadays yellow disease has been spread widely and very harm, so the preventif actions is needed. One of the preventif actions of virus disease is through healthy seed. The aims of this research were to know the health of commercial long bean seed, the occurence of virus on seed from diseased plant, and the type of virus which infected long bean. The methods used on this research were by visual observation of seed, growing-on test, molecular detection, protein analysis and observation of virus morphology using electron microscope. Seed cultivars used in this research were New Jaliteng, Java Plus, Mahkota 01, Hijau Super, Brigade and collection seed from Plant Virology Laboratory of Gadjah Mada University. The result revealed that seed from a pack of commercial seed showed malformation and didnt have uniformity in size. The collection seed from plant with yellow symptoms showed malformation and wrinkled symptoms. The result on growing-on test method showed leaves with mosaic, yellowing, stunted, curly and malformation symptoms, but molecular detection and protein analysis of BCMV and Begomovirus didnt show the DNA band or protein band which indicated virus attack. Long bean seed which was tested and showed symptoms on growing-on test revealed that it didnt bring neither BCMV or Begomovirus. This research also proved that Begomovirus on long bean wasnt transmitted by produced seed.
