The purpose of this study was to understand species diversity and fatty acid profile of Order Perciformes fishes in Sukamade river, Meru Betiri National Park, East Java. Purposive sampling method was used in obtaining the sample, using hand net as a collecting tool. Sample identification was done in Animal Systematics Laboratory of Biology using identification key book Freshwater Fishes of Western Indonesia and Sulawesi (Kottelat and Whitten, 1993). Petroleum eter was used to extract the fat and fat methilation was done by using BF3 in 20% methanol. Methyl ester was separated from fatty acid by gas chromatography. Morfological feature described and used to distinguish the species. Morphologic, morphometric and meristic data were convered into quantitative data and used in building dendograms by UPGMA using MVSP progam. Essential fatty acid composition of each species were then compared to each other. The identification shows that there are 9 familys, 13 genera and 13 species in the sample obtained. The diversity of Perciformes Order fishes of Sukamade River, Meru Betiri National Park is considered to be relatively low. Species with highest abundance are: Acentrogobius janthinopterus, Rhyochichthys aspro, Eleotris melanosoma and Kuhlia marginata. Four dominant species have varied fatty acid amount and compositions.
