Summary:Plaque is the main etiology of periodontal disease, of which include a chronic periodontitis. The growth of Fusobacterium nucleatum as plaque former could cause severe chronic periodontitis. Prevention of periodontal disease severity can be done by eliminating and preventing of plaque formation using a natural product that has antibacterial capabilities, one of which is papaya leaf which contains alkaloids, flavonoids, saponins, steroids, tannins and triterpenoids. The aim of this study was to determine the effect of papaya leaf extract on the growth of plaque bacteria Fusobacterium nucleatum ATCC 25586 in vitro. This study was using 25 samples that divided into 5 groups, which consist of 3 treatment groups (papaya leaf extract 2,5%, 5%, 10%), a negative control group (papaya leaf extract 0%), and a positive control group (chlorhexidine gluconate 0,2%). Those solutes were dropped 50 μL into 5 wells on 5 TSA petri dishes that had been swabbed with bacterial suspension Fusobacterium nucleatum. The diameter of inhibition zone formed was measured 3 times (diagonal, horizontal, and vertical) using a sliding caliper to the nearest 0,01 mm and then the mean was calculated. The data obtained then analyzed by One Way ANOVA and LSD0,05 tests. The result of One Way ANOVA test showed that there was an effect of papaya leaf extract against plaque bacteria growth inhibition zone Fusobacterium nucleatum ATCC 25586. The result of LSD0,05 test showed a significant difference (p < 0,05) in bacteria growth inhibition zone among groups of papaya leaf extract concentration). The widest inhibition zone was on the concentration of 2,5% and then was decrease on the concentration of 5% and 10% (inhibition zone 2,5% > 5% > 10%). The conclusion from this study was that the papaya leaf extract could inhibit the growth of plaque bacteria Fusobacterium nucleatum ATCC 25586.
