Summary: | Porphyromonas gingivalis is one of the bacteria that cause periodontal
disease. Red betel leaf contains antibacterial compounds. There are two
processing method of red betel leafs in the community that the efectivity hasnt
been proven yet.
The aim of this study is to determine the inhibition of Porphyromonas
gingivalis bacteria after application of 10 and 20 pieces of red betel leaves that
boiling and steeping with aquades. Porphyromonas gingivalis was used a pure
culture with the consentration of 108CFU/ml. The effect of 10 and 20 pieces
boiling water and steeping water of red betel leaf (Piper crocatum) to the growth
of Porphyromonas gingivalis was determined by measuring the optical density
(OD) of Porphyromonas gingivalis using spectrophotometre with 600nm
wavelength. The initial OD was measured before incubation, while the latter OD
was measured after incubation for 24 hours. The final OD was obtained from the
latter OD minus the initial OD.
This study showed that the application of boiling water and steeping water
of red betel leaf (Piper crocatum) with the number of 10 and 20 pieces does not
give a significantly effect to the growth of Porphyromonas gingivalis. This result
was probably caused by the thickness of the membrane structure of
Porphyromonas gingivalis bacteria and the mechanism of protection, acted by the
outer membrane and periplasmic and caused by also the lack of active
antibacterial compounds of red betel leaf (Piper crocatum) with 10 and 20 pieces
of leaves. |
