ETD Background : Kembang bulan [Tithonia diversifolia (Hemsley) A. Gray] has been studied has many benefits, one of them as an anticancer drug candidate. In vivo studies on the effect of ethanol extract standardized leaf flower in the function and liver histopathology has not been done before. Objectives : Standardize the ethanol extract of T.diversifolia with marker Tagitinin C and study the effect of standardized ethanol extract of T.diversifolia on liver function and histopathological picture of Swiss strain mice. Methods : This research used a quasi experimental study design conducted in 60 Swiss strain mice, 30 males and 30 females. Kembang bulan leave extract was standardized with TLC method. Mice were divided into 6 groups with doses of 2000, 1000, 500, 250, 125 mg/kgBB standarized ethanol extract of kembang bulan and 1 mL of water in the control group. Provision made orally for 14 days. On day 14 liver function test was performed and mice were sacrificed for liver histopathologic examination. Results : Kembang bulan leave extract has active compund of tagitinin C with Rf 0,32 in 2:1 comparison (of wash benzene:ethyl acetate). The result of liver function showed that there were no increase of SGOT and SGPT levels that exceed the normal range. Histopathological examination showed mild inflammation in the liver of 1 female mice in group I (2000 mg/kgBB), IV (250 mg/kgBB), and V (125 mg/ kgBB), mild coagulation necrosis with inflammation in the liver of 1 female mice in group I (2000 mg/kgBB) and III (500 mg/kgBB), and severe coagulation necrosis in the liver of 1 female mice in group V (125 mg/kgBB). The changes in histopathological picture that occurs in mice is not affected by increasing the dose and is more influenced by the condition of each individual mice. Conclusion : Ethanol extract of kembang bulan leave can be standarized by tagitinin C. Standarized ethanol extract of kembang bulan up to a dose of 2000 mg/kgBB did not have any impact on liver function and and at a dose of 125 mg/kgBB caused mild inflammation and severe coagulation necrosis with inflammation, 250 mg/kgBB caused mild inflammation, 500 mg/kgBB caused mild coagulation necrosis with inflammation, and the dose of 2000 mg/kgBB causes mild inflammation and mild coagulation necrosis with inflammation.
