Si., Apt. ETD Background: Kembang bulan [Tithonia diversifolia (Hemsley) A. Gray] is one of plants which widely used as traditional medicine and has potentiality as anticancer drug. Ethanol extract (70%) of Kembang Bulan�s leaves contains terpenoid compounds and has a cytotoxic effect to WiDR cells with IC50 = 53,19 μg/mL, stronger compared to cytotoxic effect of 5-Fluorouracil (5-FU) which has IC50 = 62,16 μg/mL. A research about its antiproliferation effect to WiDR cell is needed to support further development of this extract as a colorectal cancer drug. Objectives: To standardize the ethanol extract of T. Diversifolia leaves using Tagitinin C as an active compound marker and find out its antiproliferation effect to WiDR cell compared to 5-FU using doubling time as parameter. Methods: This quasi-experimental research using WiDR cell as research subject. Standardization of T. Diversifolia leaves ethanol extract was done by Thin Layer Chromatography method to reveal Tagitinin C as its active compound. Each sample was made into 8 serial doses for standardized T. Diversifolia leaves ethanol extract (0,78125-100 μg/mL) and 5-FU (0,9375-120 μg/mL), given to WiDR cell and incubated at 37oC for 0, 24, 48, and 72 hours. Proliferation rate was measured by colorimetric enzyme activity using MTT method and the absorbance rate was read using ELISA reader. Antiproliferation effect to WiDR cell was determined using doubling time as parameter. Results: T. Diversifolia leaves ethanol extract was standardized using Tagitinin C as active compound marker with Rf value = 0,32. The best antiproliferation effect of this extract to WiDR cell was found at dose of 1/64 IC50 = 0,78125 μg/mL with 174,1 hours of doubling time, longer than the longest of WiDR cell doubling time which treated by 5-FU, 142,8 hours at dose of 1/2 IC50 = 30 μg/mL. Conclusion: Antiproliferation effect of standardized T. Diversifolia leaves ethanol extract to WiDR cell is dose-dependant. It�s increasing along with decreasing of the dose respectively.
