Biosynthesis of Ag nanoparticle utilizing natural reducing agent becomes an alternative method for biomedical application. This research focus on the influence of Piper betle leaf extract on Ag nanoparticle stability and their antibacterial test on pathogenic bacteria E. coli and S. aureus. by chemical reduction The synthesis of Ag nanoparticle was conducted method using chitosan as stabilizer and Piper betle leaf extract as reduction agent obtained by liquid-liquid extraction. AgNO3 solution was added into 1% solution of chitosan continued with addition of the extract and heated at the 100 ºC for 2 hours. Starting concentration of AgNO3 was varied from 100, 200, 300, 400 and 500 ppm. The antibacterial activity was tested using disk diffusion method. P Based on GC-MS analysis result, iper betle leaf contains derivative compound of phenol (4-allyphenyl acetate, eugenol, eugenyl acetate, chavicol and allylpyrocatechol diacetate) which has a role in reducing Ag + into Ag 0 . Produced Ag nanoparticle has a spherical shape in the size of 25-35 nm for 50 ppm concentration and 24-72 nm for 250 ppm concentration. Ag nanoparticle has fcc structure with lattice parameter of 4.088 �. Antibacterial test result show that bacteria inhibition index increased along with the increase of Ag nanoparticles concentration.
