Vincristine and vinblastine are antimitotic compounds contained in Tapak Dara (Catharanthus roseus [ L ] G. Don .) has been studied to produce melon (Cucumis melo L.) cultivars melody Gama - 1 become autotetraploid. This study aimed to determine the effect of immersion time variation of ethanolic extract of Tapak Dara leaf to phenotype characters and ploidy melon (Cucumis melo L.) cultivar Hikadi. Two-day old sprouts melon soaked with ethanolic extract of Tapak Dara leaf with a concentration of 0.05 % by long immersion 6 , 12 , 18 and 24 hours. Then transferred to polybags and etched with ethanolic extract of Tapak Dara leaf on the apical bud. 7 days old melon seedlings transferred to pots in green house KP4 and harvested at 60 days after planting. Phenotypic characters examined included plant height, stem diameter, number of leaves, fresh weight of fruit, round fruit, thick flesh, thick skin, horizontal diameter of fruit, vertical diameter of fruit, number of seeds per fruit, weight of 100 grams of seeds, seed length, broad beans, seeds and thick. Chromosome number of root tip cells of plants sprout melon on the 2nd generation is used to determine the degree ploidy of melon results from soaking treatment ethanolic extract of Tapak Dara leaf. The results obtained in this study obtained soaking treatment ethanolic extract of Tapak Dara leaf with a concentration of 0.05 % with 12 hours soaking time results phenotype of plants with characters who are superior to others. This is evidenced by the character of the habitus of plants and fruits that are superior to other treatments or controls soaking time. At microscopy observations on some cell plant sprouts on the 2nd generation, the observed total number of chromosomes (2n=48).
