Summary:Panulirus homarus is one of the high economically valued spiny lobster which is mostly caught and cultured at south coast of Lombok Island among the other spiny lobsters. The purposes of this reasearch are to observe the genetic variations within Panulirus homarus populations in Lombok by Inter-Simple Sequence Repeats (ISSR) markers, and to determine which population in Lombok is the most potential for juvenile harvesting, parents selection, dan conservation. Sampling was done in three locations, which are Gerupuk Bay, Bumbang Bay and Awang Bay. DNA amplification was done by PCR-ISSR method using ISSR1 and ISSR3 primers. Data analysis was done by UPGMA method to construct a dendrogram. Morphological characters of body width/length ratio and coloration do not show significant differences between the three populations. There were total of 48 polymorphic loci amplified from the samples of three analyzed populations. Population in Gerupuk Bay has the highest genetic variations in the ISSR regions among the three analyzed populations based on their polymorphism percentages and similarity values. Branching pattern of the dendrogram shows the clustering of 23 analyzed samples into three distinct groups consistent with the three sampling locations.
