Summary:One species of brown algae potential producer of alginate is Sargassum. In the making of alginate, water-soluble polysaccharides such as fucoidan and laminaran are dissipated as waste. Fucoidan has been used but laminaran has not been in efficient utilization, so to conduct a research is essential to find its potential. The general objective of this study was to obtain laminaran prebiotic of brown algae S.crassifolium. The specific objectives of this study were (1) to obtain an efficient laminaran extraction method and physico-chemical, chemical and functional characteristics of laminaran crude extract (2) to obtain the enzymatic hydrolysis or depolymerization time of laminaran crude extract producing laminaran oligosaccharides with DP less than equal 9 (bifidogenic), (3) to determine the effect of treatment process and gastric juice to chemical stability of laminaran prebiotic oligosaccharides, (4) to determine the index of laminaran prebiotic oligosaccharides compared with standard laminaran and commercial prebiotics (inulin) and (5) to determine the ability of laminaran oligosaccharides in producing SCFA on fermentation in vitro using bacteria from feces. The best extraction method was the method 5 (Yvin modification) that algae powder was extracted with acid filtrate asulfat then precipitated with ethanol followed by reextraction of algae residue with distilled water and the resulting filtrate was also precipitated with ethanol. The extraction of brown algae S. crassifolium using Yvin modification method produced two types of extracts i.e. is laminaran extract lysis with acid so that the result still contained many impurities from the cell wall components its names LAE (Laminaran Acid Extraction) and laminaran extracts resulted from the washing of algae residue with distilled water so that its main content was LME (Laminaran Modified Extraction).
