Summary:The objective of this research was to determine the potential of polyphenols extract and phlorotannins extract of Padina pavonica in inhibiting α-amylase and α- glucosidase. Polyphenols was extracted using 50% methanol, while phlorotannins was extracted using 96% methanol and non-lipid layer was separated using distilled water, methanol, and chloroform, then partitioned using ethyl acetate twice. Total content of polyphenols and phlorotannins were tested, then analyzed using TLC. Phlorotannins extract was analyzed further using HPLC. Both types of compounds were tested to know their ability to inhibit α-amylase and α-glucosidase activity. Total phenolic content of Padina pavonica was 17.63±0.25 µg GAE/mg extract and total phlorotannins content was 1.94±0.19 µg PGE/mg extract. The results showed that polyphenols and phlorotannins can inhibit α-amylase (IC50 polyphenols = 357.26±54.82 µg/ml and IC50 phlorotannins = 251.80±22.14 µg/ml), but lower than acarbose (IC50 = 61.12±7.22 µg/ml). Polyphenols and phlorotannins can also inhibit α-glucosidase (IC50 polyphenols = 26.57±2.08 mg/ml and IC50 phlorotannins = 34.40±5.93), but lower than acarbose (IC50 = 1.74±0.25 mg/ml). Both compounds have higher activity for inhibiting α- amylase activity than α-glucosidase. There was high correlation between total phenolic and phlorotannins with inhibition percentage of α-amylase and α-glucosidase. HPLC results showed that Padina pavonica phlorotannins extract contained of phloroglucinol.
