Summary:One way to improve rice production, the most important cash crop in Indonesia, is increasing the potential of superior cultivars. New plant type (NPT) rice, introduced by IRRI, is an ideotipe which is characterized by medium tiller number (8-10 tillers) but all are productive. This characteristic is now being introduced to �Ciherang�, one of the most popular cultivar in Indonesia, and is expected to raise spikelet number per panicle. The grand strategy chosen is by developing Chomosome-segment Subtitution Lines (CSSL) population, based on 'Ciherang' with NTR donors, namely 'Fatmawati', B12743, and B11143D. As a preliminary work, this study aimed to select the polymorphic microsatellite markers based on 'Ciherang' and the NTR donors. The selected markers then were used to verify F1 individuals, whether the hybrids were true heterozygots. All the parental materials were observed morphologically and genotyped. Observation was conducted on flowering time (20 %, 50 %, and 80 %, calculated in days after sowing), plant height, flag leaf width, flag leaf length, flag leaf area, tiller number, productive tiller number, panicle length, spikelet number per panicle, and 1000 grain weight. Survey on polymorphism was conducted on 513 microsatellite markers, selected for their equal distribution among rice 12 chromosomes. The result showed that the morphological differences found between cultivars yield components 'Ciherang' with lines B11143D are on flowering time, plant height, flag leaf length, flag leaf area, tiller number, productive tiller number, panicle length, spikelet number per panicle, and 1000 grain weight. This study found 155 polymorphic microsatellite markers, spread over 12 chromosomes among cultivars 'Ciherang' and lines 'B11143D' which cover 86,15% of the chromosomes in average. Based on molecular verification, based on RM6909, RM405 and RM20582-Xa7 markers, we verified that 26 of 27 F1 plants tested were heterozygous.
