ETD Indonesia is a country with great potential for natural products, especially medicinal plants. The rhizome of Curcuma zedoaria (Berg.) Roscoe as one of nutritious plants, has been used empirically in the communities for treatment. The variation of growing area influences environmental conditions that are likely to influence the content of compounds in plants. This study aims to determine the values of parameter nonspecific, specific and content of the eugenol compound in the extracts of zedoary rhizome as well as the effect of variation of growing area for the level of eugenol on standardized extract zedoary rhizome from Kalibawang, Tuksono and Tawangmangu to produce zedoary rhizome with the highest levels of eugenol. The dried powders of zedoary rhizome are macerated with 96% ethanol after choiced the optimum solvent. Extract is standardized and determined the levels of eugenol. The levels of eugenol in the extract compared with the effect of variation of growing area likes altitude of growing area, light intensity, pH and soil moisture. The results are compared by ANOVA and t-test with a level of significance was 95%. The results showed that the standardization of extract is fulfilled the requirement excepts the total ash content and the levels of eugenol extracts were significantly different from Kalibawang, Tuksono and Tawangmangu. The altitude of growing area, light intensity, pH, and soil moisture had a significant effect on the level of eugenol for ethanol extracts. Tuksono was the area that produces zedoary rhizome with the highest levels of eugenol which was 0.18%.
