Summary:BACKGROUND: Ischemic stroke is a heterogeneous disease with various underlying pathophysiology. Genetic factor plays a crucial role among non-modifiable risk factors. OBJECTIVE: This research aims to identify leptin gene polymorphism as a risk factor for insulin resistance and ischemic stroke. METHOD: This research consists of a total of 84 samples, 42 cases and 42 controls. Case refers to blood sample of ischemic stroke patient while control refers to blood samples of non-ischemic stroke patient with no history of stroke. Blood samples of all subjects underwent DNA isolation and Polymerase Chain Reaction (PCR) and electrophoresis were done and visualized by staining with ethidium bromide and viewed under ultraviolet (UV) rays to study the polymorphism of microsatellite allele of Leptin gene in these samples. RESULTS: Results showed that there is no direct correlation between leptin gene polymorphism, incidence of ischemic stroke and insulin resistance.
