ETD Early flowering can be induced by genetic transformation through the insertion of flowering gene of rice Heading date 3a (Hd3a) gene that controlled by strong promoter of CaMV (35S pro) to overexpress this gene in P. amabilis. Candidate of P. amabilis transformant that contained T-DNA 35S::Hd3a has been obtained using hygromycin containing New Phalaenopsis selection medium. Positive transformants were analyzed by both phenotypic and molecular approach. P. amabilis orchid transformant candidates were examining by PCR technique using HPT primers (HygF1 primer and HygR1 primer) and Hd3a gene specific primers (35SO primer and Hd3aR primers). To determine the effect of Hd3a gene on development of P. amabilis, morphological observation were carried out in 3 � 10 months old plants in the number of leaves, number of roots, length and width of leaves and plant height. Phenotypic analysis showed that 35S::Hd3a transformed plants were slower grow and produced different shoot growth habit compare to that of non transformants (wildtype) plants. About 1000 basepairs amplified T-DNA 35S::Hd3a fragment from the genome DNA of transformants indicate that T-DNA has been integrated to the genome of P. amabilis. This results suggest that Hd3a gene as inserted and integrated fragmen into the genome of P. amabilis caused the change in shooting and growth speed of shoot of 35S::Hd3a orchid transformants.
