PATOGENISITAS FUSAN Bacillus thuringiensis var. kurstaki DAN B.t.var. israelensis BIAKAN AIR KELAPA PADA ULAT KUBIS Crocidolomia binotalis ZELL.

The use of coconut water as alternative based culture medium to fusan of B.t. (F28, F31, and F33) was investigated. The coconut water optimimize to obtain cell mass, find out their pathogenicity, and also sub letal effect to C. binotalis. This research revealed that fusan well grown in coconut water...

(F28, F31, and F33) was investigated. The coconut water optimimize to obtain cell mass, find out their pathogenicity, and also sub letal effect to C. binotalis. This research revealed that fusan well grown in coconut water and produce a protein crystal. The fusan cell mass of whole coconut culture after 4 days cultured is F28 (9,75 x 109 cells/ml), F31 (3,49 x 109 cells/ml), and F33 (6,5 x 109 cells/ml). Pathogenicity of F28 higher than F31 and F33 to C. binotalis larvae. LC value of F28 to second instar larvae are LC50 (4,99 x 108 cells) and LC90 (1,09 x 109 cells) whereas third instar larvae is LC50 (7,91 x 108 cells) and LC90 (3,27 x 109 cells). Second instar of larva, F31 has LC50 (1,25 x 109 cells) and LC90 (1,06 x 1011 cells) but third instar larvae LC50 (5,27 x 1015 cells) and LC90 (4,77 x 1022 cells). F33 has LC50 (1,03 x 1017 cells), LC90 (2,14 x 1017 cells) to second instar larvae and third instar larvae LC50 (1,84 x 1018 cells) dan LC90 (2,98 x 1025 cells). Second instar of C. binotalis larvae more susceptible than third instar larvae. Exposured of fusan also cause sub letal effect such as decelerate of larvaes growth and development, decrease of pupae and imago size, failed inclusion or death of pupae, abnormal form of imago and pupae. Hence, coconut water based culture medium is effective for growth and pathogenicity of fusan (F28) to C. binotalis.
