Summary:Broody behavior is closely related to the synthesis of the prolactin hormone, and prolactin promoter gene polymorphism. The aims of this research were to study the relation between the broody behavior of the Hen (from F1 to F4) with the polymorphism of prolactin promoter gene and inheritance of the broody behavior to the offspring. This study was doing in October 2013 - March 2014 at University Farm (KP4 UGM) Berbah, Sleman and Laboratory of Genetics of Faculty of Biology UGM. The research was using 1 chicken F1 White-Spots, F1 White-Brown, F1 Striped, F3 and F4. Hens were kept in semi-intensive cage at KP4 UGM. At the time of egg-laying period, the hens broody behavior was observed and the sample of blood was take as much as 3 mL. DNA was isolated using the protocol of ROCHE®. DNA from blood samples were amplified using PCR-RFLP (Restriction Fragment Length Polymorphism). Restriction enzyme that was used in this research was HindIII, digested at 37oC with incubation tie for 1, 4, 8, 12, 16, 20, and 24 hours. The result show that the negative expression of broody behavior in F1 generation found just in F1 Striped, while that trait can be inherited from F3 to its offspring F4. The result of electrophoresis from the sample after digestion with restriction enzyme HindIII did not show any polymorphism in the prolactin promoter gene. Conclusion of this research is the not-broody behavior trait can be found in GAMA Chicken F1 Striped, F3, F4 and can be inherited to the offspring, and the polymorphism of promoter prolactin gene in all of sample GAMA Chicken was not found.
