Summary:Tooth extraction causes wound formation at the site of extraction. Wound healing process consists of inflammation, proliferation and tissue remodeling phase. Sea cucumber (Stichopus noctivagus) contains flavonoid and saponin which act as an anti-inflammation and antibacterial in the healing process. This study aimed to determine the effect of 75% sea cucumber extract gel towards the infiltrations of polymorphonuclear leucocyte and macrophage cells. This study involved 16 guinea pigs (cavia cobaya) aged 3-4 months old. The subjects were randomly divided into 2 groups namely treatment and control groups. Tooth extraction was performed on the lower left incisors. Twenty microliter 75% percent sea cucumber extract gel was applied into the socket of the treatment group, while 2% carboxymethyl cellulose was applied into the socket of the control group. The guinea pigs were then decapitated on 1, 3, 7, and 14 days after the treatment. The specimens were embedded in paraffin, sectioned serially, and stained using hematoxylin eosin. Polymorphonuclear leucocytes and macrophages were observed under microscope. ANOVA showed significant differences (p<0.05) among the groups, indicating that 75% sea cucumber extract gel affected PMN leucocyte and macrophage infiltration. The number of PMN leucocyte and macrophage infiltration of the treatment group was significantly (p<0.05)less than the control group in all observation periods. In conclusion, application of 75% sea cucumber extract gel into post extraction socket wound reduce the number of PMN leucocyte and macrophage infiltration in the post extraction wound healing process.
