Summary:Tarsius sp is one of endemic primate in Indonesia that is threatened. One of the efforts to preserve Tarsius sp is conservation. Tarsius bancanus and Tarsius sp. from Sulawesi morphologically are difficult to differentiate, so they need genetic study. The objective of this research is to know the genetic diversity in Tarsius bancanus borneanus, Tarsius bancanus bancanus dan Tarsius dianae and their classification based on NADH dehydrogenase 1 (ND1) encoding gene. Sampling and DNA isolation of T. b. borneanus, T. b. bancanus and Tarsius dianae. Amplification of ND1 gene by PCR technique with forward ND1 primer and reverse ND1 primer. The product of PCR is 1278 bp DNA segments which then were processed by sequencing. The result of ND1 gene sequences (957 bp) were then been multiple aligned with other primates which were taken from Genbank using MEGA version 5.1. The result of analysis showed that there were 160 sites of different nucleotides and 33 sites of different amino acids. Genetic distances based on nucleotide sequence of ND1 gene were analyzed using Kimura 2-parameter. The genetic distances between T. b. bancanus and T. b. borneanus is 0,4%, between T. b. bancanus and T. dianae is 19,8% and between T. b. borneanus and T. dianae is 19,7%. Neighbor-Joining method was used to construct the phylogenetic trees based on nucleotide sequence and amino acid sequence of ND1 gene with bootstrap 1000 times. Based on the phylogenetic trees there is genetic diversity between T. bancanus and T. dianae (from Sulawesi), but there is no genetic diversity between T. b. bancanus (from Sumatera) and T. b. borneanus (from Kalimantan). NADH dehydrogenase 1 gene sequence could be used to classify Tarsius in subordo Strepsirrhini.
