Identification based on physical appearance was done by analyzing the differences on the hair colour and pattern, size, habitat, and vocalization. Its complex distribution pattern makes these species and subspecies difficult to be distinguished. Because of that, a research to determine the species of tarsier is necessary to conduct. The purpose of this research is to analyze the specific genetic diversity between Tarsius bancanus borneanus, Tarsius bancanus bancanus, and Tarsius spectrum based on the encoding gene sequence of NADH Dehydrogenase Sub-unit 1 (ND1). This research employed tarsiers DNA isolated from the muscles of Tarsius bancanus borneanus, Tarsius bancanus bancanus, and Tarsius spectrum. PCR amplification with ND1 Forward primer and ND1 Reverse primer resulted in (produced/showed) amplicon with 1278 bp in length. Besides, the data analysis using MEGA 5.1 Beta 4 and multiple alignment using Clustal W showed a nucleotide sequence of ND1 encoding gene with 957 nt in length. The resulted sequenced gene of ND1 was compared to other primates from Genbank. The result showed that 164 sites of nucleotide and 28 sites of amino acid can be used as the genetic markers between Tarsius bancanus and Tarsius spectrum. The average genetic distance of tarsier based on the nucleotide sequence of ND1 coding gene analyzed using Kimura 2-parameter was 11,5%. Phylogenetic tree constructed using Neighbour joining method with bootstrap value of 1000 times could differentiate Tarsius bancanus and Tarsius spectrum, but it could not differentiate between Tarsius bancanus borneanus and Tarsius bancanus bancanus.
